Returning head coach the Ireland coach has named captain Doris in his Ireland roster for the autumn internationals, however lock McCarthy is absent.
The captain, absent from play since having a shoulder operation in the start of May, recently returned to provincial training, but it remains unclear if he will be fit to play against the All Blacks in the US on 1 November.
Lock McCarthy, on the other hand, did not make the cut after failing to recover from the foot injury suffered during the first Lions match against the Wallabies in the summer.
Ulster player Cormac Izuchukwu and Munster winger Calvin Nash also miss out due to injury, and full-back Keenan had already been ruled out after having a hip operation after the summer tour.
Leinster forward McCarthy - the brother of Joe - is the only new player in the 34-player group, but the Irish Rugby Football Union says uncapped provincial duo Edogbo and Gleeson and once-capped Connacht winger Shayne Bolton will head to the US as "backup".
Following facing the All Blacks at Soldier Field in a repeat of their 2016 encounter, Ireland will play Japan, Australia and world title holders the Springboks in Dublin.
"There's an exciting look to the panel and there's an opportunity for the more experienced players and for the newcomers on the summer tour to the Lelos and the Lobos," commented Farrell, back to national team duties after a break to lead the Lions tour to the southern hemisphere.
In a largely familiar-looking squad, Leinster's Prendergast and the southern club's Jack Crowley will once again vie for the first-choice number 10 position. The number 10 jersey is remains up for grabs, with Prendergast and Crowley poised to face off for the first time in Saturday's URC match at Croke Park.
Frawley is the third fly-half chosen as the coach deciding not to calling up Leinster's Byrne.
At number 9, Farrell has picked Lions tourist Jamison Gibson-Park and Casey, who captained the national side to the summer international wins over Georgia and the Lobos. There is also a recall for Connacht's Blade, chosen to club-mate Murphy.
Henshaw, Aki, Garry Ringrose and McCloskey represent an veteran group of midfield choices. Jamie Osborne is a further choice in the centres, though the youngster is also a leading candidate to deputise for Keenan at full-back.
Outside backs Lowe, Mack Hansen, O'Brien and Stockdale are named. The winger impressed on his national team debut during the summer with two tries apiece in the victories over the Lelos and Portugal.
Paddy McCarthy is the sole new name in the Ireland pack. The front rower has started a couple of his province's URC United Rugby Championship matches so far and featured in Ireland A's defeat by England in the English city previously.
The 22-year-old was included in interim boss O'Connell's summer selection, yet did not earn a cap.
Southern province lock Thomas Ahern, who earned his debut appearances in those Tests, is kept in the squad, and veteran Ulster lock Henderson returns after missing the summer tour.
Jack Conan, who played in all three Lions matches over the summer, is set to deputise for the captain if the skipper does not regain fitness in time to play the All Blacks.
The Ireland squad will assemble in Dublin on Monday before traveling to Chicago on Tuesday.
